Yeast

Saccharomyces cerevisiae is a powerful model for understanding genetic regulation in eukaryotic cells. Dharmacon yeast resources include multiple collections for interrogating the yeast genome, including Tagged ORFs, Kockout Strains, Protein-Protein Interaction Collections and Mutant Strains and Screening Collections.

Tagged ORFs

• Yeast ORFs

  Individual gateway-adapted yeast ORFs from the Yeast ORF collection.

• Yeast ORF Collection

  The Yeast ORF collection consists of Gateway-adapted yeast ORFs cloned into expression plasmids. ORFs can be expressed as fusion proteins or moved to other vectors.

• Yeast YFP Fusion Kinase Collection

  The Yeast Kinase YFP Fusion collection contains a number of kinase genes that have been cloned with a C-terminal YFP tag and promoter region into a Gateway-entry vector.

  The Molecular Barcoded Yeast (MoBY) ORF library was designed to identify genes in Saccharomyces cerevisiae that confer drug resistance when mutated, providing a tool for identifying drug-resistant mutations in yeast.

  The Yeast Cross and Capture system is designed to examine protein-protein interactions and protein modifications using 6xHIS and VSV tagged haploid yeast strains.

E. coli

Includes the E. coli Keio knockout collection, Promoter-GFP fusions for monitoring gene expression, and tagged ORFs for studying protein-protein interactions. Escherichia coli has long been a model organism for molecular genetics, with the first experiments observing bacterial conjugation occurring in the 1940's. E. coli products include collections useful for further understanding gene regulation, protein-protein interactions, and analysis of mutational effects on gene function.

• E. coli Keio Knockouts

  The E. coli Keio Knockout Collection was designed to provide researchers with in-frame, single-gene deletion mutants for all non-essential E. coli K-12 genes.

• E. coli Promoter Collection

  The E. coli Promoter Collection is a library of fluorescent reporter strains in which green fluorescent protein (GFP) is used to measure promoter activity in Escherichia coli.

• E. coli Tagged ORFs

  E. coli Tagged ORFs include both SPA- and TAP-tagged open reading frames intended to facilitate the mapping of protein-protein interactions in Escherichia coli.

C. elegans

C. elegans ORFs, RNAi feeding collections, promoter and transcription factor collections. The nematode Caenorhabditis elegans is an important model organism for biological research, and has lead to breakthroughs in the areas of apoptosis and RNA interference (RNAi).

• C. elegans ORF Collection The C .elegans ORF Collection, created as a resource to functionally characterize the Caenorhabditis elegans proteome, consists of a collection of predicted protein-coding open reading frames (ORFs) that have been cloned into a Gateway-donor vector.
• C. elegans Promoter Collection Designed for construction of promoter-fusion constructs, the C. elegans Promoter Collection consists of over 6,000 Gateway-adapted promoters.
• C. elegans RNAi Collection Developed for RNAi screens in C. elegans, this RNAi collection includes full open reading frames for over 11,000 genes cloned into a feeding vector.
• C. elegans Transcription Factors C. elegans Transcription Factors have been cloned into Gateway-adapted vectors and can be used for yeast one-hybrid (Y1H) and two-hybrid (Y2H) assays.

3. Using the datafile, look up ORF ID or Clone Catalog Number of your clone of interest
   a.) for transcription factors, search by sequence name variant
4. Search by ORF ID or Clone Catalog Number in the top-right search box on the Horizon website
5. Add Clone to your Cart and proceed to checkout.

Horizon (formerly Dharmacon) is a distributor of multiple gene expression clone collections (cDNAs and ORFs). These clone collections were generated by groups outside of Horizon and thus the quality of the collections is largely dependent upon what Horizon received from these groups. Specific clone information and plate coordinates were provided to Horizon by the suppliers of these clone collections. Horizon has not sequence verified each individual clone from these collections. These collections and individual clones are distributed "as is" with no additional product validation or guarantees. Horizon has established quality procedures to ensure that individual clones are picked from the identified well in a plate, grown on the correct antibiotic, and are free of phage contamination. Due to the quality of the information provided to Horizon , the clone you receive might not match the expected clone. If this occurs, please contact Technical Support.

All clones and plates are provided as glycerol stocks. Individual strains are shipped at room temperature. Plates are shipped on dry ice. Both should be stored at -80ºC.
