Pre-arrayed gene families of expression-ready ORFs

Gene over-expression studies are one of the most common tools used in genomics to study gene function in mammalian cells. Clones of gene Open Reading Frames, or ORFs, cover the initiating methionine to the last amino acid and lack the 3' and 5' UTRs found in cDNA clones. ORFs are used in functional studies ranging from whole genome over-expression screening to protein localization to RNAi validation, and many other applications. However, the availability of expression-ready resources has often been limited and pre-arrayed gene families have not always been readily available. This has led to unnecessary limitations on gene function studies. To address this, we have created a whole new set of gene family libraries within the CCSB-Broad Lentiviral Expression ORF collection.

There are now pre-arrayed libraries for a large number of common gene families including: phosphatases, kinases, proteases, GPCRs, and many more! There is also the option to select from two different genome-scale resources, a druggable genome library or the complete collection. These new libraries open up several new areas of research to gene over-expression studies. New gene family libraries make it much easier for you, the researcher, to get exactly the ORF reagents you need to focus your experimental plan. Large-scale or small-scale, you will find just the right resource to advance your work!

CCSB-Broad Lentiviral ORF highlights

  • CCSB-Broad Lentiviral Expression Clones (OHS6085) are fully sequenced, and match their accessions with no more than one silent mutation (no amino acid change) within the open reading frame.
  • Lentiviral expression vector – allows creation of lentiviral particles for transduction of almost any mammalian cell type.
  • Can also be transfected for expression right from the plasmid.
  • CMV promoter drives strong expression of the ORF construct
  • Blasticidin resistance allows for multiplex experiments in cells containing Puromycin or Neomycin resistance.
  • V5 tag facilitates pull-downs and co-immunoprecipitation experiments in order to determine protein-protein interactions, or detection by fluorescent antibodies.
  • Delivered as a glycerol stock for an infinite resource.
