SMARTvector Inducible RNAiコントロールは、遺伝子特異的なSMARTvector Inducible Lentiviral shRNA実験の前に、最適なデリバリー条件を決定するための費用対効果の高いツールです。shMIMIC Inducible Lentiviral microRNAのコントロールとしても使用できます。

遺伝子特異的なコンストラクトのSMARTvector Inducible Lentiviral shRNAまたはshMIMIC Inducible Lentiviral microRNAを注文する際には、目的の遺伝子を検索し、ご希望のプロモーターとレポーターを選択します。

SMARTvector Inducible RNAiコントロールは、遺伝子特異的SMARTvector Inducible Lentiviral shRNAおよびshMIMIC InducibleLentiviral microRNAを使用する前に、デリバリーおよびドキシサイクリン誘導の最適条件を決定するためのコスト効率の高いツールです。

初めに、SMARTchoice Inducible Non-targeting Control 4-Packを使用して、細胞に最適なベクター構成を特定してください。

SMARTchoice Inducible Non-targeting Control 4-Pack（カタログ番号：VSC6847）を使用して、4つのプロモーターを同時に評価することにより、ご使用の細胞において最もアクティブなプロモーターを決定します。

SMARTvector Inducible non-targetingコントロール 1：Horizonで最も長く、最も一般的に使用されているSMARTvector non-targetingコントロールです。ほとんどの実験をサポートするのに理想的です。

SMARTvector Inducible Lentiviral shRNA実験ワークフローは、SMARTchoice Inducible Non-targeting Control 4-Packを使用して、形質導入細胞のTurboGFP蛍光強度を評価し、目的の細胞において最もアクティブな RNAポリメラーゼIIプロモーターを特定することから始まります。次に、遺伝子特異的な SMARTvector Inducible Lentiviral shRNAおよび、それと同一のベクター構成（同じRNAポリメラーゼIIプロモーターとレポーター）を持つポジティブおよびネガティブコントロールを注文します。ドキシサイクリンの用量（濃度）および誘導時間の最適化は、ポジティブコントロールとネガティブコントロールを使用して行う必要があります。目的の遺伝子を標的とするSMARTvector Inducible Lentiviral shRNAを細胞に形質導入します。その際には、厳密なデータ解釈のために、ポジティブおよびネガティブの対応するRNAiコントロールと常に同時にアッセイをすることが必要です。SMARTvector Inducible Lentiviral shRNAおよびRNAiコントロールは、即時形質導入用の高力価の精製および濃縮レンチウイルス粒子として提供されます（大腸菌グリセロールストックフォーマットも有り）。

SMARTvector Inducible Lentiviral shRNAプロモーター活性は細胞タイプによって異なる

図中の細胞株に、SMARTvector Inducible Lentiviral shRNA Non-targetingコントロールをMOI = 0.3で形質導入しました。形質導入の24時間後、non-targeting shRNAおよびTurboGFPの発現を1 µg/mLドキシサイクリンで誘導しました。ドキシサイクリンの存在下で48時間培養した後、細胞をHoescht-33342で染色し、核（青）とTurboGFP（緑）を画像化しました。フィールド内の細胞の約30％が形質導入されていますが、特定の細胞タイプでは、恒常的プロモーター活性が低いことを示すTurboGFP発現が低いため、一部の画像には30％未満のTurboGFP陽性細胞が含まれているように見える場合があります。

A: U2OS細胞に、non-targetingコントロールshRNA（NTC）またはユビキチンB（UBB）遺伝子に対するshRNAのいずれかを運ぶSMARTvector Inducible Lentiviral shRNAベクター（mCMVプロモーター）をMOI = 0.1で形質導入しました。1.5 µg/mLピューロマイシンで3日間細胞を選択し、選択後、細胞を96ウェルプレートに1ウェルあたり2000細胞で播種しました。24時間後（1日目）、指示された用量のドキシサイクリンでshRNA発現を誘導し、CellTiter-Glo®アッセイ（Promega）で細胞数を毎日測定しました。各データポイントは、6つの独立したウェルの平均と標準偏差を表します。
B: 6日目に、ドキシサイクリンへの5日間の曝露後、細胞をHoescht 33342で染色し、細胞核（青）とTurboGFP（緑）をArrayScan™ VT IHCSリーダーで画像化しました。
