Summary: We demonstrate in CAR-T cells multitargeting of several therapeutically relevant loci, retaining proliferative and cytotoxic ability, including advantages over Cas9. Aptameric recruitment enables simultaneous knock-in and knockout and allows targeting polygenic disorders.
