Given the relatively short-term experimental timing with synthetic reagents, how long does it take to increase transcriptional output of genes through CRISPRa?

An increase in gene expression (mRNA) can be measured as soon as 48 hours after introduction of the reagents into the cells with observed effects out to 5 days and beyond. 72 hours appears to be a consistent spot for most cells and genes. The extent of change in expression depends on the basal level of gene expression as a more dramatic increase is to be expected for genes that are expressed at lower levels and a less observable change for genes already abundantly expressed.
