To enable better understanding of the role of phosphoinositides in basic cell and disease biology Horizon has knocked out a number of key genes involved in phosphoinositide metabolism that are expressed in HAP1 cells and non-essential.
Phosphatidylinositol and Phosphoinositides are a minor component of plasma and intracellular membranes. Lipid phosphatases and kinases can target the inositol group at the level of the hydroxyls situated in the positions 3, 4, and 5 generating a total of 7 phosphoinositides.
Many of these molecules have been identified as essential mediators of signal transduction in various cellular processes, even some of those initially thought to be simple intermediates. Deregulated phosphoinositide metabolism is linked to various human diseases and cancer.
Further research is necessary to elucidate the mechanism of action of phosphoinositides and the proteins involved in their metabolism in the wide range of cellular processes and compartments were they have been found.
