What are the "up" and "down" primers listed in the gene search results for the yeast KO strains?
These are the barcode tags and can be used for verification of the strains. The barcode tags are part of the UpTag and DownTag primers listed on the Yeast Deletion Project homepage (in primer downloads section). The UpTag and DownTag primers are used in the first round of PCR to construct the yeast KO strains. These 74bp primers consist of (5' to 3'): 18 bp of genomic sequence that flank either the 5' or 3' end of the ORF (directly proximal and distal to the start and stop codons respectively), 18 and 17 bp of sequence common to all gene disruptions.
