Video: How to measure viral titer using fluorescent colony counting
Viral titers are determined in HEK 293 cells, which are easy to transduce. Most other cells types are transduced with lower efficiencies than HEK 293. If you are working in another cell type, you will need to determine the functional titer of your lentiviral preparation relative the HEK 293 titer listed on the side of the tube to determine functional titer specific to your cell type. This brief video walks you through the steps of functional titer determination using a lentiviral vector expressing a GFP reporter, a fluorescent microscope and the colony counting method.
