Horizon offers both western blotting and cell-based ELISAs to analyze activation of signal transduction pathways and to measure changes in protein expression. Western blots offer a qualitative method for assessing protein expression and modification (phosphorylation, ubitquitination etc.) in response to alterations in cell signalling pathways. We can offer independent investigation of nuclear and cytoplasmic extracts. Cell-based ELISAs quantitatively investigate the expression of specific proteins, for example to help to identify a compound’s mechanism of action.
PI3K and ERK pathway activation in X-MAN® HCT116 cells containing PI3K and KRAS mutations. HCT116 colorectal carcinoma cells were grown overnight in normal or low serum conditions and whole cell lysates were analysed by western blotting using antibodies to components of the PI3K and KRAS pathways.
The ability of the PI3K/mTOR inhibitor BEZ-235 to inhibit phosphorylation of the downstream protein S6. MCF7 breast cancer cells were treated with BEZ-235 for 2 h and phosphorylation of S6 ribosomal protein measured by ELISA.
